Zehn Jahre nach den Ereignissen in Raccoon City wird Agent Chris Redfield nach Afrika geschickt.
Chris ist einer der Gründungsmitglieder der BSAA (Bioterrorism Security Assessment Alliance) und auch dessen bester Agent.
In Afrika angekommen wird Chris von einer Frau angesprochen - Sheva Allomor, seine neue Partnerin.
Sheva ist Afrikanerin, die der BSAA nur beigetreten ist weil ihre Eltern durch Bio - Terrorismus umgekommen sind.
Gemeinsam begeben sich die beiden auf die Jagt nach einem Mann namens Irving, der der einigste Anhaltspunkt ist.

Je weiter die beiden kommen umso mehr erfahren sie über die Wahrheit der Ereignisse in Afrika.
Nachdem die beiden ihre erste Mission erfüllt haben - Sicherstellung von Geheimen Dateien die aus der Forschungseinrichtung kopiert wurde - entdeckt Chris ein Bild...
Auf diesem ist seine, für tot erklärte, alte Partnerin Jill Valentine zu sehen.
Plötzlich scheint ein Funken Hoffnung in Chris aufzulodern das Jill evtl. doch noch am Leben sein könnte da ihre Leiche niemals gefunden wurde...
